The Nooksack dace (Rhinichthys cataractae) is a small cyprinid fish listed as Endangered under Canada's Species at Risk Act due to urban, industrial, and agricultural development and associated loss of riffle habitat. We tested multi-marker environmental DNA (eDNA) metabarcoding for detecting Nooksack dace and co-occurring fish communities at thirteen sites within its fragmented distribution in southwestern British Columbia. Twelve native and ten non-native fish species were detected across two markers and two sampling years. Nooksack dace detections within the Nooksack River watershed were generally consistent with historical data; they were not detected in the Brunette River or other potentially suitable areas sampled. Results suggest eDNA metabarcoding is an effective, high-sensitivity tool for monitoring Nooksack dace populations and assessing species distribution and recovery. The detection of several provincial priority invasive species demonstrates the value of eDNA metabarcoding for simultaneously supporting species at risk monitoring and invasive species surveillance. This approach can assist conservation and recovery efforts for Nooksack dace by improving our understanding of their distribution, population status, and ecological interactions.\n\n

eDNA samples were collected from ten sites in the Lower Fraser Valley, British Columbia. The sites included tributaries of the Nooksack River (Bertrand Creek and two of its tributaries, Fishtrap Creek, and Pepin Creek) as well as tributaries within the Little Campbell River watershed, representing a range of urban land-use settings in southern British Columbia. In October 2024, nine of the sites sampled in 2022 were resampled, and additional sampling locations were added. These included Fergus Creek (a tributary of the Little Campbell River), the Brunette River, and the Sumas River, both tributaries of the Fraser River.","sampling":"Water sampling and filtration were conducted using an eDNA Backpack Sampler with 5 µm self-preserving filters (Smith-Root, Inc.). To minimize contamination, multiple cleaning steps were performed before sampling at each site. A field blank was collected at each site prior to sample collection by opening a 1.0 L bottle of Millipore water for approximately one minute and subsequently filtering it. In both sampling years, up to nine water samples were collected per site, with a target volume of 3 L per sample. The number of samples collected varied among sites due to factors such as stream flow conditions and filter clogging. Following collection, filters were stored at room temperature according to the manufacturer’s recommendations until eDNA extraction.
