Se cortaron cuidadosamente muestras de hojas, tallos y tronco.","methodSteps":["Se realizó la colecta manual del tejido vegetal proveniente de especies de árbol de cacao variedad FSV41-2. Se tomaron muestras de hojas y tallos de los árboles, que se guardaron hasta su procesamiento en laboratorio.","Aislamiento de cepas de hongos endófitos: En una cabina de flujo laminar se cortaron cuadrados de 0.5 cm de lado tanto de las hojas como de corteza. De las hojas se cortaron tres (3) trozos al azar y se clasificaron como a, b y c, siguiendo la metodología propuesta por Hoyos et al. (2023).","Los trozos de material se lavaron con solución Tween 80 (0.01%), agua y etanol (70%), durante un minuto por cada solución. Después se sumergieron en hipoclorito de sodio (3%) por 15 min. Luego, se repetió el ciclo con Tween 80 y agua. Finalmente, las muestras se sembraron en cajas Petri con agar PDA a pH 4.5, suplementado con clindamicina (0.2 mL por cada 100 mL). El período de incubación duró 7 días a 28°C. En caso de presentarse crecimiento de más de un hongo, se repicó cada uno por separado en medio PDA. Las cepas asiladas destinadas a conservación serán preservadas en medio PDA con aceite mineral estéril siguiendo el protocolo descrito en Long-Term Preservation of Fungal Cultures in All-Russian Collection of Microorganisms (VKM): Protocols and Results, de Ozerkaya et al. (2013), con algunas modificaciones.","Extracción de ADN, identificación molecular y fenotípica: Se extrajo el ADN fúngico usando el kit de ADN de tejido EZNA® (Omega Bio-Tek).","Luego, se cuantificó el ADN total usando un espectrofotómetro NanoDrop ND-1000 (NanoDrop, Wilmington, DE). Se emplearon cebadores fúngicos universales para amplificar la región del ADN ribosómico del espaciador transcrito interno (ITS1) (5'-AGAGTTTGATCHYTYAGATGG‐3' y 5'-TTGTTAACCACGGYTCGACTT‐3') por PCR y haciendo uso de secuenciación automática de secuencia de tinte.","Las PCR se realizaron a 94 °C durante 1 min, y 35 ciclos de 94 °C: 30 s, cebador específico Tm 60°C durante 30 s, y 72°C durante 5 min, utilizando la ADN polimerasa Taq (Thermo Fisher Scientific) en un termociclador Swift™ MiniPro (ESCO). Los productos de PCR se visualizan en un gel de agarosa al 1% y se purificaron usando Wizard SV Gel y PCR Clean-Up System (Promega, EE. UU.) antes de secuenciarse en un secuenciador ABI prism 3500 (Applied Biosystems). Se analizaron las secuencias de ADN y se compararán con las disponibles en GenBank a través de la búsqueda BLAST (https://blast.ncbi.nlm.nih.gov/Blast.cgi).","Adicionalmente, se realizó la caracterización microscópica de las estructuras reproductivas y vegetativas, empleando el método de impresión con cinta adhesiva haciendo tinción con azul lactofenol (Merck®) y KOH (Merck®).","La cepa se cultivó en cuatro medios de cultivos convencionales y en un medio diseñado bajo limitación nutricional con el cual se busca potenciar la producción de metabolitos bioactivos. Los medios convencionales serán: YM (10 g/L extracto de malta, 4 g/L D-glucosa, 4 g/L extracto de levadura, preparado en agua desionizada; pH 6.3), Q6 ½ (2.5 g/L D-glucosa, 10 g/L glicerina, 5 g/L harina de semilla de algodón, preparado en agua desionizada; pH 7.2), ZM ½ (5 g/L melaza, 5g/L avena, 4 g/L sacarosa, 4 g/L manitol, 1.5 g/L D-glucosa, 1.5 g/L carbonato de calcio CaCO3, 0.5 g/L edamina, 0.5 g/L sulfato de amonio (NH4)2SO4, preparado en agua desionizada ; pH 7.2), y un medio sólido de arroz BRFT (cada unidad se prepara con 28 g de arroz integral, 1 g/L extracto de levadura, 0.5 g/L tartrato disódico dihidratado, 0.5 g/L fosfato monopotásico KH2PO4 y 100 mL de agua desionizada). Todos los cultivos se harán en Erlenmeyers de 500 mL y se incubarán a 23ºC.","El nuevo medio se diseñó bajo limitación nutricional, buscando causar estrés metabólico en el hongo. Se tendrá en cuenta el perfil molecular generado en los medios convencionales con el fin encontrar un medio que permita potenciar la producción de compuestos bioactivos.","Una vez crecida la cepa en los diferentes medios, se hará un proceso de extracción con solventes orgánicos. Para los medios líquidos, el micelio y el sobrenadante se separan por filtración. Al micelio se le agrega acetona y se lleva a un baño ultrasónico por 40 minutos a 40ºC. Luego, se filtra nuevamente y se evapora la acetona hasta obtener únicamente agua. Posteriormente, a la fase acuosa obtenida se le agrega acetato de etilo y se agita vigorosamente. El acetato de etilo se recupera, se filtra usando sulfato de sodio y se lleva a secar al vacío usando un rotaevaporador. Por otro lado, para la extracción del sobrenadante, únicamente se agrega acetato de etilo y se hace el procedimiento anteriormente mencionado. Con respecto al medio de arroz (medio BRFT) se hace el proceso de extracción que se realizó para el micelio de los cultivos líquidos. El proceso de extracción del cultivo crecido bajo el nuevo medio diseñado se determinará según el crecimiento del hongo en el sustrato proporcionado.","Una vez obtenidos los extractos crudos, estos serán analizados por cromatografía líquida acoplada a masas (HPLC/MS). En donde a partir de los resultados se determinará cuáles medios de cultivos permiten obtener compuestos bioactivos de interés. Los extractos crudos que se compongan de posibles moléculas antimicrobianas o de compuestos novedosos se purificarán usando técnicas de cromatografía líquida (HPLC) para aislar las moléculas que componen el extracto.","Las moléculas aisladas a partir de los diferentes cultivos del hongo serán identificadas por medio de resonancia magnética nuclear (NMR) y se les harán pruebas de citotoxicidad y de concentración mínima inhibitoria (MIC) contra diversas cepas patógenas con el fin de identificar su actividad antimicrobiana.","La metodología presentada se basa en el trabajo de diversos autores, como lo son: Charria-Girón et al., 2023; Hassan et al., 2022; Harms et al., 2021; Kemkuignou et al., 2022; Khruengsai et al., 2023; Llanos-López et al., 2023; Mitschke et al., 2023."]},"countryCoverage":[],"collections":[],"dataDescriptions":[],"dataLanguage":"spa","pubDate":"2026-09-24T00:00:00.000+00:00","maintenanceUpdateFrequency":"AS_NEEDED","maintenanceDescription":"","license":"http://creativecommons.org/licenses/by-nc/4.0/legalcode","dwca":{"coreType":"http://rs.tdwg.org/dwc/terms/Occurrence","extensions":["http://data.ggbn.org/schemas/ggbn/terms/Permit"],"modified":"2026-09-24T18:17:54.614+00:00"},"approximateCounts":{"occurrenceCount":45}}
