The records contained here include eDNA metabarcoding results from the 12S (targeting fish) and COI (targeting general biodiversity) markers. At each of five coastal sampling stations we conduct beach seining to catch and measure fish, and collect water samples for eDNA metabarcoding. Beach seining and eDNA can complement one another, since neither method will capture every species found at the sampling sites on their own. ","language":"eng","citation":{"text":"Jeffery N, Stanley R, Pettitt-Wade H (2026). At each of the sites, we first collect water samples by boat from over top of the eelgrass beds. Three samples of 3 litres of water are collected and filtered per sample site. Samples are filtered with a Smith-Root portable vacuum pump on to a 1.2 micron PES filter to capture eDNA. At each site, a field blank consisting of 1-2 litres of distilled water are also filtered to check for potential contamination. These filters are frozen at -80 degrees Celsius until processing by the lab. At each site, we also conduct two beach seine hauls by boat, to collect fish and invertebrates in the eelgrass meadows. All animals collected are processed in the field and held in aerated totes of seawater. Invertebrates are identified but not counted, and all fish are identified and counted. A subset of 50-100 individuals per fish species are measured (total length) to obtain size distributions for each species. \n\nIn the lab, eDNA samples are extracted using commercial kits, and libraries for Illumina sequencing are created using primers for each marker of choice (e.g., MiFish-U for 12S targeting fish and Leray primers for COI targeting general eukaryotic biodiversity). These libraries are sequenced on an Illumina NovaSeq or MiSeq platform. Raw eDNA reads are demultiplexed and processed using the QIIME2 pipeline, with denoising and sequence merging conducted by DADA2. Amplicon sequence variant tables are created, and representative DNA sequences are assigned taxonomy using nucleotide BLAST against public reference databases.","methodSteps":["First, sample sites are chosen. Amplicon sequence variants (ASV) tables are generated by DADA2 for each ASV per sample site, and taxonomy is assigned to each ASV using nucleotide BLAST against the NCBI eukaryotic nucleotide database. The taxonomy is then filtered for relevant taxa (i.e., marine fishes and invertebrates) and manually curated to remove spurious observations."]},"countryCoverage":[],"collections":[],"dataDescriptions":[],"dataLanguage":"eng","acknowledgements":"This work was funded by the Marine Conservation Targets program with the department of Fisheries and Oceans Canada. ","pubDate":"2026-02-19T00:00:00.000+00:00","maintenanceUpdateFrequency":"ANNUALLY","maintenanceDescription":"Samples are collected from these coastal sites annually.
